FEBS Practical and Lecture Course

FEBS Practical and Lecture Course - A summer school aimed at graduate students, post-docs, and young independent investigators who want to learn about new techniques to explore the proteins encoded by the human genome.The course will teach state-of-the-art analysis of the human proteins with a focus on antibody-based methods, transcriptomics and mass spectrometry.
The course will have several daily lectures by experts in the field and this will be complemented with workshops in which the participants will annotate genes/proteins. Each participant will take part in an annotation effort with the aim to go through all experimental evidence of the respective protein and to evaluate the quality of the underlying data to score the evidence for protein profiles and functionality.

ACTIP 30th anniversary meeting

Mathias Uhlen will hold the keynote presentation "Generation of a resource of the human secretome proteins by animal cell culture and implications for drug development" on May 6 at 16.30 pm.

Dr. Jan Mulder will present "An integrated omics approach provides a detailed view on the molecular landscape of the brain" on Thursday October 10, 17:50, at the VIB Conference series "The Brain Mosaic: Cellular heterogeneity in the CNS (2nd edition)" held in Leuven, Belgium.

The brain is the most complex organ of the mammalian body with a diverse physiology combined with a complex cellular organization. To further expand our basic understanding of the neurobiology of the brain and its diseases, there is a need for a comprehensive, in-depth molecular dissection of the various brain regions and cell types. In the Human Protein Atlas (HPA) project, we use an integrated omics approach based on multiple transcriptomics methods and antibody-based mapping to generate regional expression profiles in the human, porcine and mouse brain. Genes with an elevated expression in the brain, or enriched in discrete regions have been classified. Our main focus is to identify the shared molecular organization of the mammalian brain compared to human specific features and to link genes to cells and brain functions.

A new open access HPA Brain Atlas as part of the Human Protein Atlas is launched (September 2019) to allow exploration and comparison of the expression of individual protein coding genes in the various parts of the mammalian brain.

WEBINAR | Addressing the reproducibility crisis: Validating antibodies for life science research

Antibodies are invaluable tools in the life sciences. There are several challenges that must be overcome to validate the specificity and reproducibility of antibody reagents, as emphasized by many publications, including a recent proposal from the International Working Group for Antibody Validation (IWGAV). There are many examples of antibodies with off-target binding leading to erroneous conclusions, some of which will be discussed during this webcast, demonstrating the importance of application-specific validation. Five alternative strategies for enhanced validation of antibodies will be described based on genetic validation, recombinant expression, independent antibodies, orthogonal validation, and capture mass spectrometry validation. Approaches based on these five strategies have been used for streamlined, enhanced validation of more than 10,000 antibodies from more than 30 providers, targeting over 7,000 human proteins as part of the Human Protein Atlas program. The results show a path forward for validating antibodies in an application-specific manner suitable for both providers and users.

During the webinar, viewers will learn about:

• The generation and validation of data used in the Human Protein Atlas
• Problems related to the cross-reactivity of antibodies and the challenges they create for the interpretation of results
• Enhanced validation of antibodies using five application-specific strategies.

The course will focus on antibody based technologies and describe the possibilities of using these technologies within the facilities at SciLifeLab. Lectures will cover both theoretical and more practical aspects of the different technologies, including the following:

• Immunohistochemistry

• Tissue fluorescence profiling
• Protein array technologies and suspension bead technologies
• Mass cytometry and flow cytometry
• Immuno capture/immuno precipitation in combination with mass spectrometry

Apart from general aspects of the technologies, the course will include project presentations to demonstrate the benefits and possibilities of the technologies in different contexts. Last but not least validation strategies of antibodies for different applications will be covered, as this is of outmost importance for adequate output data.
